    Slow and Fast Neocortical Oscillations in the Senescence-Accelerated Mouse Model SAMP8

    The senescence-accelerated mouse prone 8 (SAMP8) model is characterized by accelerated, progressive cognitive decline as well as Alzheimer's disease (AD)-like neurodegenerative changes, and resembles the etiology of multicausal, sporadic late-onset/age-related AD in humans. Our aim was to find whether these AD-like pathological features, together with the cognitive deficits present in the SAMP8 strain, are accompanied by disturbances in cortical network activity with respect to control mice (SAM resistance 1, SAMR1) and, if so, how the alterations in cortical activity progress with age. For this purpose, we characterized the extracellular spontaneous oscillatory activity in different regions of the cerebral cortex of SAMP8 and SAMR1 mice under ketamine anesthesia at 5 and 7 months of age. Under these conditions, slow oscillations and fast rhythms generated in the cortical network were recorded and different parameters of these oscillations were quantified and compared between SAMP8 and their control, SAMR1 mice. The average frequency of slow oscillations in SAMP8 mice was decreased with respect to the control mice at both studied ages. An elongation of the silent periods or Down states was behind the decreased slow oscillatory frequency while the duration of active or Up states remained stable. SAMP8 mice also presented increased cycle variability and reduced high frequency components during Down states. During Up states, the power peak in the gamma range was displaced towards lower frequencies in all the cortical areas of SAMP8 with respect to control mice suggesting that the spectral profile of SAMP8 animals is shifted towards lower frequencies. This shift is reminiscent to one of the principal hallmarks of electroencephalography (EEG) abnormalities in patients with Alzheimer's disease, and adds evidence in support of the suitability of the SAMP8 mouse as a model of this disease. Although some of the differences between SAMP8 and control mice were emphasized with age, the evolution of the studied parameters as SAMR1 mice got older indicates that the SAMR1 phenotype tends to converge with that of SAMP8 animals. To our knowledge, this is the first systematic characterization of the cortical slow and fast rhythms in the SAMP8 strain and it provides useful insights about the cellular and synaptic mechanisms underlying the reported alterations

    Undergraduate Kinesiology Student Involvement in the Department and Sense of Belonging by Employment Status

    Many undergraduate college students find the need to work either on or off campus in order to pay for their tuition and expenses. It is unknown whether or not employment impacts a students ability to be involved in the department or feel like they belong and not typically reported for Kinesiology programs. PURPOSE: To assess the impact employment had on college students in relation to their sense of belonging and involvement with their home department and to investigate belonging and involvement in Kinesiology students. METHODS: Undergraduate college/university students (18+ years) at a public institution in Southern California were recruited via flyers, social media, announcements and word of mouth to participate in an IRB approved online research study. The cross-sectional Qualtrics survey consisted of a few demographic questions (age, gender, major, employment status, etc.), 9 questions specific to involvement in the department (6-point Likert scale: 1=strongly disagree to 6=strongly agree) and 11 questions for belonging divided into two domains; social acceptance and valued competence (same Likert scale as involvement). For the involvement and belonging questions an average and sum score were determined for each individual, which was used in the statistical analysis. For employment, participants were asked if they were employed (yes/no) during the Spring 2023 semester, which was then coded to form the groups. Statistical analysis software (IBM SPSS v.28) was used to assess the differences between those that indicated they were employed and not employed and a p-value of 0.05 was implemented for significance. RESULTS: College undergraduate participants (N=149, age; 24.3 ĺśź 5.7 years, gender identification; n=1 other, n=111 female, and n=37 male, and Kinesiology n=83, or other majors n=59) indicated they were employed (n=86) or unemployed (n=26) and Kinesiology students reported similar employment statistics (employed n=51 and unemployed n=11). An Independent T-Test found no significant differences (P\u3e0.05) in involvement or belonging for employment status in all students and for those who declared Kinesiology as their major during Spring 2023. CONCLUSION: Employment status had no significant impact on an undergraduate student\u27s involvement in their major department or their sense of belonging

    Identification of a novel sequence motif recognised by the ankyrin-repeat domain of zDHHC17/13 S-acyl-transferases

    S-acylation is a major post-translational modification affecting several cellular processes and being particularly important for neuronal functions. This modification is catalysed by a family of transmembrane S-acyl-transferases that contain a conserved zinc-finger DHHC (zDHHC) domain. Typically, eukaryote genomes encode for 7-24 distinct zDHHC enzymes, with 2 members also harbouring an ankyrin-repeat (AR) domain at their cytosolic N-terminus. The AR domain of zDHHC enzymes is predicted to engage in numerous interactions, and facilitates both substrate recruitment and S-acylation-independent functions; however, the sequence/structural features recognised by this module remain unknown. The two mammalian AR-containing S-acyltransferases are the Golgi-localised zDHHC17 and zDHHC13, also known as Huntingtin-interacting proteins 14 and 14-like, respectively; these are highly expressed in brain, and their loss in mice leads to neuropathological deficits that are reminiscent of Huntington disease. Here, we report that zDHHC17 and zDHHC13 recognise via their AR domain, evolutionary conserved and closely related sequences of a [VIAP][VIT]xxQP consensus in SNAP25, SNAP23, Cysteine-String Protein, Huntingtin, Cytoplasmic Linker Protein 3 and Microtubule Associated Protein 6. This novel AR-binding sequence motif is found in regions predicted to be unstructured, and is present in a number of zDHHC17 substrates and zDHHC17/13-interacting S-acylated proteins. This is the first study to identify a motif recognised by AR-containing zDHHCs

    El objetivo general de la investigación fue determinar la relación entre la gestión educativa y el clima organizacional en los Centros de Educación Técnico Productiva del Callao, 2020. La población estaba compuesta por 30 docentes de los Centros de Educación Técnico Productiva del Callao, 2020. La metodología usada fue el hipotético-deductivo, el diseño de estudio es no experimental de nivel correlacional de corte transversal; se recolectó la información en un momento específico, la técnica usada fue el de la encuesta y el instrumento de recolección de datos el cuestionario. Para la confiabilidad del instrumento se utilizó el alfa de Cronbach que salió alta en ambas variables: 0,979 para la variable gestión educativa y 0, 967 para la variable clima organizacional. Para los resultados descriptivos se hizo a través de las tablas de distribución de frecuencias y el gráfico de barras. Y para la contrastación de la hipótesis se realizó a través del coeficiente de Rho de Spearman. El estudio concluyó estableciendo que existe una correlación buena (Rh0= 0,701) entre la gestión educativa y el clima organizacional en los Centros de Educación Técnico Productiva del Callao, 2020. Siendo P valor =0,000 <0,05; lo cual indica que existe correlación. Esto afirma la hipótesis y el objetivo general del estudio.The general objective of the research was to determine the relationship between educational management and the organizational climate in the Centers for Technical Productive Education of Callao, 2020. The population consisted of 30 teachers from the Centers for Technical Productive Education of Callao, 2020. The The methodology used was hypothetical-deductive, the study design is non-experimental with a cross-sectional correlational level; The information was collected at a specific time, the technique used was that of the survey and the data collection instrument the questionnaire. For the reliability of the instrument, Cronbach's alpha was used, which was high in both variables: 0.979 for the educational management variable and 0.967 for the organizational climate variable. For the descriptive results, it was done through the frequency distribution tables and the bar graph. And to test the hypothesis, it was performed through Spearman's Rho coefficient. The study concluded by establishing that there is a good correlation (Rh0 = 0.701) between educational management and organizational climate in the Technical Productive Education Centers of Callao, 2020. Being P value = 0.000 <0.05; which indicates that there is a correlation. This affirms the hypothesis and the general objective of the study

    Total reflection X-ray fluorescence analysis using polycapillaries. A comparison with conventional setups

    The possibility of producing parallel X-ray beams with low divergence by means of half monolithic polycapillaries gives the impression to be useful in total reflection X-ray fluorescence (TXRF) experiments. On one hand, the use of polycapillaries facilitates the alignment and the setup of the experiment. As expected, the spectra registered in the experiment shown low background and good signal-noise ratio. On the other hand, the intensity of photons on the samples when polycapillaries are employed is lower than in other configurations, which produces a loss of efficiency for the excitation of the sample mainly for light elements. In this work, different TXRF experiments were carried out and the minimum detection limits attained were compared with the ones obtained from TXRF using polycapillaries. The results indicate that the decrease of intensity produced by polycapillaries is imposing when detection limits are analyzed. Nevertheless, detection limits are better than conventional XRF. The possibility of employing non-symmetrical polycapillaries could eventually improve the detection limits so as to be equivalent to the conventional TXRF setups. © 2010 Elsevier B.V.Fil: Sanchez, Hector Jorge.     A computational model coupling mechanics and electrophysiology in spinal cord injury

    Traumatic brain injury and spinal cord injury have recently been put under the spotlight as major causes of death and disability in the developed world. Despite the important ongoing experimental and modeling campaigns aimed at understanding the mechanics of tissue and cell damage typically observed in such events, the differenti- ated roles of strain, stress and their corresponding loading rates on the damage level itself remain unclear. More specif- ically, the direct relations between brain and spinal cord tis- sue or cell damage, and electrophysiological functions are still to be unraveled. Whereas mechanical modeling efforts are focusing mainly on stress distribution and mechanistic- based damage criteria, simulated function-based damage cri- teria are still missing. Here, we propose a new multiscale model of myelinated axon associating electrophysiological impairment to structural damage as a function of strain and strain rate. This multiscale approach provides a new framework for damage evaluation directly relating neuron mechanics and electrophysiological properties, thus provid- ing a link between mechanical trauma and subsequent func- tional deficits

    Análisis de la perspectiva de la fotografía boudoir como arte contemporáneo en Lima Perú, 2022

    La presente investigación tuvo como objetivo central analizar la influencia del estereotipo en la fotografía boudoir como arte contemporáneo en la ciudad de Lima. La investigación es cualitativa, básica, con diseño fenomenológico; se entrevistó a 4 especialistas. Los resultados obtenidos indicaron que efectivamente la fotografía boudoir es hoy un género fotográfico que busca transmitir la belleza de un cuerpo femenino a través de una imagen donde la mujer está dentro de su propia comodidad, este arte es confundido con la fotografía erótica y esto causa que este estilo no sea nombrado y reconocido por lo que transmite sino por lo que provoca, y es ahí cuando la sociedad confunde este género con un estilo de fotografía inapropiado para ser difundido. Con estos resultados se concluye que la fotografía boudoir es hoy un arte de libre expresión donde se aprecia el cuerpo como un lienzo de pintura que transmite lo que verdaderamente la mujer siente, teniendo como una de sus finalidades sentirse bella y feliz consigo misma

    Direct characterization of functional materials by haptenized fluorescent dendrimers for in vitro allergic drug diagnose

    β-lactams are the most widely drug prescribed against infections, but they are the most commonly reported medication allergy too. A correct diagnosis of these allergic reactions is crucial to avoid rejecting them by other more expensive broad-spectrum antibiotics, with potential risk factors for the development of multiple drug-resistant bacteria. [1] Skin testing is the most consensual approach to diagnose β-lactam allergy, but this in vivo test is not risky free and should be performed under strict hospital surveillance.[2] In vitro testing is not still widely used on account of their low sensitivity. We report the use of already haptenized fluorescent dendrimers [3] to control the preparation of materials for in vitro test, and their verification by testing on patient sera samples. This fluorescent dendrimer is obtained from a dye with two orthogonal functional groups suitable for click chemistry.
